Effects of I¥êB and Bcl-2 overexpression on cell death in mouse cortical collecting duct cells

Abstract
Recent data have implicated nuclear factor ¥êB (NF¥êB) and Bcl-2 in the prevention of apoptosis in various cell lines. However, it has been also reported that NF-¥êB plays an important role in inflammatory responses in renal epithelial cells and that effect of Bcl-2 in necrotic cell death is not clear. We investigated the effect of specific inhibition of NF¥êB and overexpression of Bcl-2 on oxidant or cisplatin-induced injury of renal collecting duct cells. M-1 cells, which were derived from outer cortical collecting duct, were stably transfected with the dominant negative mutant inhibitor of NF¥êB (NF¥êB¥áM) and Bcl-2. I¥êB¥áM significantly decreased oxidant-induced injury, but had ono significant effects on cisplatin-induced necrotic cell death. However, I¥êB¥áM sensitized cells to apoptosis induced by cisplatin or serum deprivation. Overexpression of Bcl-2 inhibited the apoptotic cell death induced by serum diprivation, treatment of cisplatin and contact inhibition, but did not affect oxidant-induced necrotic cell injury in collecting duct cells. These data show that activation of NF¥êB mediates oxidants-induced necrotic injury, but inhibits apoptotic cell death in renal collecting duct cells.
